These are quite interesting designs; however, I believe most are somewhat difficult to synthesize – especially at the speeds we require.
I’ll post an example route to DAR-DIA-033-2, as an example.
This initial substructure should be doable by this urea formation reaction 
We we see that EN300-708755 can take part in “formation of urea” for an easy synthesis.
Next the algorithm would have us perform a Negishi; however, I don’t believe either the Negishi or the preparation of the organozinc would be particularly quick to perform. 
We can certainly check with Enamine. I will also comment on the other molecules when I wake up tomorrow!
