Comparison of IC50 values for MAT-POS-bb423b95-7 and ADA-UCB-6c2cb422-1 suggests that methylation of the amide nitrogen in the 3-amino series will result in a signficant reduction in potency. This is likely to result from a shift in the cis/trans geometrical preference of the amide as discussed here.
